Matrixyl 3000

A palmitoyl peptide blend where two different numbers get conflated — and one of them is more overstated than it first appears. The peer-reviewed trial behind the underlying peptide was funded by a rival manufacturer and used a concentration a thousand times lower than the figure usually quoted for it. The manufacturer's own flagship figure, separately, measures wrinkle area, not depth.

In brief

Should you care? If fine lines and early collagen loss are your concern, yes — but two things about the underlying evidence are commonly overstated. A placebo-controlled trial of the underlying palmitoyl peptide found improved wrinkle depth by objective measurement after 12 weeks1 — but it was funded by Procter & Gamble, a competing manufacturer, and used a concentration of 3 parts per million, not the 3–5% sometimes quoted. The manufacturer's own headline figure for the full Matrixyl 3000 blend — a striking 45% — measures something different again: wrinkle area, not depth. See below for why both distinctions matter.

What it actually is

Matrixyl 3000 is a trade name — not a single molecule but a blend of two fragments, palmitoyl tripeptide-1 and palmitoyl tetrapeptide-7, developed and sold by a single cosmetic ingredient manufacturer. The "palmitoyl" part is a fatty-acid tail attached to help the peptide penetrate skin, since peptides on their own are typically too large to cross the skin barrier well.

Because it is a proprietary branded blend rather than a generic compound, most of the published testing on it comes from the manufacturer itself, which is worth knowing before treating the numbers as fully independent. What the evidence supports

Two distinct results get quoted here, and they are often conflated — including, previously, on this page. A placebo-controlled, split-face trial of palmitoyl pentapeptide (Pal-KTTKS — the same fragment family as Matrixyl 3000's palmitoyl tripeptide-1) in 93 women found improved wrinkle depth by objective optical measurement after 12 weeks of twice-daily use.1 Two things about that trial are easy to miss: it tested a concentration of 3 parts per million — roughly a thousand times lower than the "3–5%" figure sometimes quoted for this research — and it was designed and funded by Procter & Gamble, a competing cosmetic ingredient manufacturer, not an independent academic group. Separately, Sederma's own proprietary technical data for the complete Matrixyl 3000 blend, at 3% concentration, reports a 45% reduction in the surface area occupied by deep wrinkles and roughly a 20% increase in skin tonicity, after two months.2 Foundational laboratory work on the isolated pentapeptide fragment shows it stimulates collagen types I and III and fibronectin production in fibroblast cultures.3

Why we grade this carefully

Wrinkle area and wrinkle depth are not the same measurement, and the 45% figure most marketing quotes is Sederma's own area result at 3% concentration — not the peer-reviewed depth result, which comes from a trial run at a fraction of that concentration and funded by a rival manufacturer. Neither number is fabricated, but both get repeated as if they describe the same, independently-confirmed thing, and neither is quite what most marketing implies. We think you should know which number you're looking at, at what concentration, and who paid for the study — which is also why we corrected our own earlier description of this trial as "independently published."

How long it takes

Sederma's own testing window was eight weeks; the independent peer-reviewed trial ran twelve. Either way, expect at least eight to twelve weeks of consistent twice-daily use before judging whether it is doing anything for you. Earlier improvements in texture and hydration are commonly reported but are not the metric either wrinkle study measured.

What it does not do

Claims the evidence does not carry

  • Work like retinol. It has a real but shallower body of evidence than retinoids, which have decades of independent trials behind them.
  • Work at any concentration. The tested figure is 3%. Many products use far less and simply list "Matrixyl 3000" on the label without a percentage — that tells you nothing about whether you are getting a tested dose.
  • Replace sunscreen or a full routine. Same caveat as every peptide on this site.

What to look for

  • A stated percentage. "Contains Matrixyl 3000" without a number tells you almost nothing.
  • Independent or third-party testing alongside the manufacturer's own data, where a brand has commissioned it.
  • A batch certificate of analysis, as with any active you are paying a premium for.
